【Function and Use】 Used for detecting smooth Brucella antibodies in bovine serum.

【Usage and Determination】

1 Usage

1.1 Sample Processing

Collect whole blood from the animal,separate the serum, and ensure the serum is clear and free of hemolysis.

1.2 Preparation of 1× Washing Solution

Before use, bring 20×Washing Solution toroom temperature (20～25℃ ), and shake to dissolve any crystals (can

be heated in a 37℃ water bath for 5～10 minutes), then dilute with purifiedwater at 1:20 , and mix thoroughly.

1.3 Dilution of Test Serum and ControlSerum

In the serum dilution plate, dilute thetest serum, negative control serum, and positive control serum with

sample diluent to 1:50.

1.4 Operating Steps

1.4.1 Sample Addition

Take the antigen-coated plate (can bedisassembled and used in batches according to the number of samples),

wash the plate once with 1× washingsolution, 300μl per well, discard the washing solution. Add the diluted test

serum, positive control serum, and negativecontrol serum to the corresponding wells of the ELISA plate, 100μl

per well. Add each test serum to 1well, andadd positive control serum and negative control serum to 2 wells

each (well 1, well 2). After sampleaddition is complete, reaction at 37℃ for 30 minutes. Remove the reaction

plate, discard the reaction solution, add1× washing solution 300μl to each well, wash 3times, and finally dry by

shaking or patting dry.

1.4.2 Add Enzyme-Labeled

Antibody Dilute HRP-labeled rabbitanti-bovine IgG with HRP -labeled rabbit anti-bovine IgG dilution at 1:200 ,

add 100μl to each well, incubate at 37℃ for30 minutes. Wash 3 times as described in 1.4.1 , and dry by shaking.

1.4.3 Color Development

Mix substrate solutions A and Bat 1:1( V:V)immediately before adding to the ELISA re-action plate, add 100μl to each well,and develop color at room temperature protected from light for 15 minutes.

1.4.4 Termination

Add stop solution 50μl to each well toterminate the reaction.
